Abstract

A demanda por enzimas nos setores industriais está aumentando nos últimos anos devido as diversas vantagens econômicas. Entre a variedade de enzimas, as proteases ganharam atenção especial nos setores industriais. Dentre as inúmeras fontes desses biocatalisadores estão os Basidiomycetes. Dessa forma, o objetivo desta pesquisa foi avaliar a produção e caracterizar as proteases sintetizadas por Pleurotus ostreatoroseus Singer. DPUA 1720. A cultura matriz, preparada em ágar batata dextrose, suplementado com 0,5% (p/p) de extrato de levedura, foi mantida por oito dias a 25 °C. Em seguida, o cogumelo foi cultivado em resíduo de mistura de Exocarpo de cupuaçu (Theobroma grandiflorum), açaí (Euterpe oleracea), abacaxi (Ananas comosus) e serragem, suplementados com farelo de arroz (Oryza sativa) ou farelo de trigo (Triticum aestivum) 0,5% (p/v) por 15 dias a 25 °C. As proteases foram extraídas em água destilada esterilizada e, no extrato bruto, foi determinada a atividade proteolítica, utilizando-se como substrato azocaseína 1 % (p/v). As proteases demonstraram as seguintes características bioquímicas: pH ótimo 5,0, temperatura ótima 40 °C, estabilidade em pH 5,0-6,0 e temperatura na faixa de 30-40 °C. A ação das enzimas proteolíticas é fortemente inibida por Fe+2 e Mn+2. Estes biocatalisadores indicam que P. ostreatoroseus tem alto potencial na indústria farmacêutica, têxtil, alimentícia e química.
