Abstract

Cogumelos do gênero Pleurotus são reconhecidos como alimentos de alto valor nutricional e por sintetizarem enzimas hidrolíticas com ampla aplicação industrial. Estas características incentivam a busca por meios que estimulem a produção de biomassa e hidrolases. Este estudo teve como objetivo avaliar meios de cultivo para crescimento, assim como, a síntese de hidrolases e a caracterização de proteases produzidas por P. ostreatoroseus DPUA 1720. A cultura estoque foi mantida em BDAYE (Ágar Batata Dextrose+Extrato de Levedura), a 25 °C por oito dias. O crescimento radial e vigor micelial foram avaliados em seis meios suplementados com Extrato de Levedura (YE): BDAYE (ágar Batata Dextrose), SABYE (Ágar Sabouraud), GYP (ágar Glicose + Peptona), MGYP (Ágar Extrato de Malte-Glicose-Peptona), ágar CEINYE ou CEDYE (Dioscorea altissima in natura ou desidratado). A avaliação da síntese de amilase, celulase e protease foi realizada em meio sólido pelo método do bloco de gelose. A influência do meio e do tempo de cultivo na produção de proteases foram realizadas por fermentação submersa. A atividade proteolítica foi determinada em solução de azocaseína 1% (p/v). As proteases produzidas por fermentação com atividade significativa foram caracterizadas quanto ao efeito do pH e da temperatura na atividade. P. ostreatoroseus apresentou crescimento rápido e micélio cotonoso em BDAYE (17,80 mm/dia) aos cinco dias de cultivo. Entretanto, SABYE e CEINYE são meios promissores para crescimento desse cogumelo. Pleurotus ostreatoroseus sintetizou quantitativo superior de amilase (17,53 mm), seguido de protease (14,33 mm) e celulase (13,61 mm) em CEINYE, BDAYE, GYP, respectivamente. Os valores significativos de atividade das proteases produzidas por fermentação submersa foram determinados em CEINYE (25,87 U/mL) e BDAYE (22,00 U/mL), aos quatro dias de cultivo. As proteases dos cultivos em CEINYE apresentaram atividade ótima em pH 5,0 e 50 °C. Pleurotus ostreatoroseus cresce, sintetiza enzimas tanto em meios de cultura tradicionais como em meios naturais e suas proteases tem potencial para aplicação na biotecnologia de alimentos.
